Do adult human oligodendrocytes remyelinate poorly and can we change this to better treat progressive multiple sclerosis?

Aim

This project covers the first aim of the project: 1) Determine whether adult human oligodendroglia in adult brain tissue in health and in MS express markers of ageing similar to those in aged rat

Compare species: rat with mouse

A preliminary comparison has been carried with published results form old-young differential expression (DE) in scRNAseq mice published by [@Ximerakis2019] and the rat differential expression from [@Neumann2019]. The DE data from the rat was not complete, it only included 31 characteristic OPC genes, 4 genes validated by qRT-PCR and 18 genes associated with senescence. This first preliminary analysis compares the genes present in one or both DE and is saved in published_rat_mouse_de

The differential expression between young and old rats from [@Neumann2019] has been redone using the raw data, as the provided data was not complete. This is in rat_young_oldDE

The comparison between mouse and rat uses the mouse from [@Ximerakis2019] differential expression and the rat DE generated earlier is available here.

Compare species: rodents and humans

A comparison with MS human brain, using the data from [@Jakel2019] is done in compare_humanMS.Rmd A comparison with HCA project, using the data from [Not publuished yet HCA] is done in rodents_HCA. This includes a DE between young human adults and old adults; that generates a list of DE genes that is compared against the genes from the DE in rodents.

Compare foetal with adult

This section uses the HCA data (not published yet) comparing the 10 foetal samples with the adult ones.
