This report summarizes analyses from a study that examines Luecosep cytokine concentrations to evalate repeatablity by time and compare Luecosep and other matrices: SepMate, CPT, and EDTA plasma to serum concentrations.
We use the Concordance Correlation Coefficient (CCC) to measure agreement of each of the plasma assays to serum. The CCC is a measure of agreement that is similar to a correlation coefficient, and has a range of -1 to 1 with \(\pm\) 1 denoting perfect agreement. We focus our evaluation of the CCC to the five analytes of interest: GM-CSF, IFN-\(\gamma\), IL-1\(\beta\), IL-6, and TNF-\(\alpha\).
For the first table (baseline over all 5 cytokines) the CCC is the “average CCC between all cytokine pairs”, we would expect a CCC of 0.85 or greater across all matrices to demonstrate “agreement” between all of them, including each matrix against serum. Ff any of the five cytokines of interest had a CCC of less than 0.85, we examined pair-wise looking at Bland-Altman plots and the scatter plots (between serum and each of the five cytokines) to find which of the five cytokines were different between serum and the other matrices.
We could do the same for For tables 2 and 3, which is the comparison on Luecosep and Serum only, although we don’t need to as the CCC is a good measure of agreement in-and-of itself because it is comparing only two matrices and any CCC < 0.8 would demonstrate agreement.
