This report tracks SPINAPS (SPN40), a study of synaptic density measured with [18F]SynVesT-1 PET in emerging and young adults with autism (with impaired or intact neurocognition) and typically developing controls. It covers recruitment, progress towards the PET targets, the data shared from the studies participants came from (SPIN-ASD, SPIN-R, SPINS) and their clinical and cognitive scores.
For questions about the methods or code, contact Hassan Abdulrasul (hassan.abdulrasul@camh.ca).
How this is counted. Each participant is counted
once, at the furthest point they have reached: enrolled (has a SPINAPS
record), eligible (Eligibility Checklist says eligible), PET done (PET
Scan Information form completed), then PET usable on QC. A PET whose QC
form is still empty is shown as QC pending rather than
unusable, because no QC decision has been made yet. Targets count usable
PETs only, because the protocol replaces participants whose PET cannot
be used. The pilot target (Aim 1) is 10 per group; the full sample (Aim
2) is 28 ASD-Impaired, 27 ASD-Intact and 29 TDC
(config/targets.csv).

| Notes. Counts are participants. PET usable = reconstructed image judged usable on QC; QC pending = PET done but the QC form is not filled in yet. Targets count usable PETs only. Not assigned = no group recorded on the Eligibility Checklist (no target). | |||||||||||
How this is counted. There is no e-consent project
for SPINAPS, so a participant is counted as recruited in the month of
their eligibility assessment (elg_doa, Eligibility
Checklist), the first study procedure after consent. Participants
without an eligibility date are left out of these plots (2 at
present).
How this is counted. A participant is withdrawn when
the Early Termination/Withdrawal Checklist says they were prematurely
withdrawn (term_premature_yn); the reason is the one ticked
on that form. This includes participants who never received a group, so
they appear under Not assigned.

What is shown. Everyone with a SPINAPS record, by group, using the SPINAPS Demographics and Height/Weight/Vitals forms. Gender is shown as recorded (check all that apply; none of the current participants ticked more than one). Mother’s education has two fields on the form and either is used. Handedness and parental education are only filled in for a few participants in SPINAPS; the From origin study tab adds them from the study each participant came from. Values entered as REDCap missing-data codes (e.g. ASKU) or outside their valid range count as missing.

What is shown. The spread of the main descriptive variables by group, for everyone with a SPINAPS record (one bin = one year of age or education; 5 IQ points). Groups are overlaid, so where they overlap the colours mix. IQ comes from the origin studies (see Data Sharing), because no IQ has been entered in SPINAPS REDCap yet.
Most SPINAPS participants took part in SPIN-ASD or SPIN-R first, and the protocol reuses their assessments and MRI instead of repeating them. This section shows where each participant’s data came from, using the Shared Data Availability form and the origin studies’ REDCap projects.
How this is counted. The studies ticked on the Shared Data Availability form, separately for clinical data and MRI. A participant who was in SPIN-ASD and then SPIN-R can have both ticked.
| Clinical data shared from | ASD-Impaired | ASD-Intact | TDC | Not assigned | Total |
|---|---|---|---|---|---|
| None recorded | 0 | 0 | 2 | 1 | 3 |
| SPIN-ASD | 7 | 0 | 5 | 0 | 12 |

Why it matters. SPINAPS does not repeat the MRI when a good-quality one exists from the referring study. Multi-shell diffusion is needed for the NODDI analyses in the exploratory aim, so it is counted separately.

How this is read. One row per participant, one column per form, from each form’s “Was this form completed?” question. Green = completed, gold = partly completed, red = recorded as not completed, grey = nothing entered. AQ and ABAS-III have no such question, so REDCap’s own form status is used: “Incomplete” is gold when the form has some answers and grey when it was never opened. The two ABAS-III forms (summary scores and questionnaire) share one column. Hover over a cell for the recorded answer and the reason a form was not completed. Not shown: SCID-5. The REDCap API returns no fields from this form, which usually means the API user has no export rights on it.
How this is read. Whether a score for each
instrument exists in the participant’s linked SPIN-ASD or SPIN-R records
(see Data Sharing), and how old it is at the SPINAPS PET.
Green = within 6 months, the protocol’s limit for
reusing an assessment; gold = older (reusable for IQ
only); grey = no origin score. Each row is labelled
with the SPN40 ID and the origin record(s) it is linked to (SPIN-ASD
SPASD_..., SPIN-R SPN3x_...). Hover over a
cell for the study, origin ID, visit and assessment date the score came
from, or, for a grey cell, which linked records were searched. This is
what could be uploaded to SPINAPS now, and what would need
re-assessing.
What is checked. Whether each ASD participant’s
recorded subgroup (Eligibility Checklist, elg_group)
matches the rule written on the same checklist. ASD-Impaired means at
least one of: MATRICS composite, RMET or Birchwood SFS total at least 1
SD below the mean of the SPIN-ASD and SPINS typically developing
controls aged 16-35; IQ < 90; or ABAS-III general adaptive composite
< 90. Otherwise ASD-Intact.
Why the checklist rule and not the protocol’s. The protocol (3.3.2) names only IQ, MATRICS and RMET; the checklist adds SFS and ABAS-III, and it is the rule staff apply. Most participants below the cut-offs are there on SFS or ABAS-III, so the difference matters.
The control norms are computed here from the SPIN-ASD control arm and the SPINS control arm (all sites), aged 16-35 at study entry:
| Measure | Controls (n) | Mean | SD | Cut-off (mean − 1 SD) |
|---|---|---|---|---|
| MATRICS composite (T-score) | 166 | 49.8 | 10.6 | 39.2 |
| RMET total | 130 | 27.9 | 4.2 | 23.7 |
| Birchwood SFS total | 181 | 172.1 | 20.5 | 151.6 |
No IQ, ABAS-III, MATRICS, RMET or SFS scores have been entered in SPINAPS REDCap yet.

Bars show the number of participants in each group with each SCID-5 diagnosis. A participant with several diagnoses is counted once for each.
Current: meets symptomatic criteria within the time window the SCID-5 specifies for the disorder (the past month for most; up to the past 2 years for some). Lifetime: has ever met diagnostic criteria (lifetime prevalence rated Threshold; in SPIN-ASD, any disorder recorded as meeting criteria). No diagnosis: no clinical disorder recorded. No current / lifetime diagnosis: at least one disorder recorded, but none meeting that tab’s definition.
From origin study: the SCID closest in time to the
participant’s SPINAPS PET among their linked SPIN-ASD and SPIN-R visits,
decoded with that study’s score sheet. The protocol reuses the SCID
history when it is older than 6 months but re-assesses current status,
so Current from an old SCID may be out of date. Decoding is the
same as in the SPIN-R dashboard
(config/scid_codes_spinr.csv,
config/scid_codes_spasd.csv).
The SCID-5 form is not exported by the REDCap API (no fields from it are returned, which usually means the API user has no export rights on it).
The SCID-5 form is not exported by the REDCap API (no fields from it are returned, which usually means the API user has no export rights on it).
Each instrument has two tabs: SPINAPS REDCap (scores entered in SPINAPS) and From origin study (the participant’s linked SPIN-ASD or SPIN-R assessment closest to their PET; see Data Sharing). Every participant is a point; hover for the participant and, in the origin tab, which record and visit the score came from and how old it was at the PET.
Self-report of autistic traits (50 items; total 0-50, higher = more traits).
No scores entered in SPINAPS REDCap yet.
Observational assessment of autism; given to ASD participants only. Calibrated severity scores (CSS, 1-10) are comparable across modules: overall, social affect (SA) and restricted and repetitive behaviours (RRB).
No scores entered in SPINAPS REDCap yet.
Clinician-rated psychopathology (18 items, each 1-7). Shown as the total and the five factor scores used in the SPIN studies.
No scores entered in SPINAPS REDCap yet.
Self-reported depression (PHQ-9, 0-27) and anxiety (GAD-7, 0-21) over the past two weeks. 5, 10 and 15 mark mild, moderate and (moderately) severe on both. These are SPINAPS-only questionnaires, so there is no origin tab content.
Not collected in the origin studies (SPINAPS only).
Full-scale IQ (standard score, mean 100, SD 15): the WAIS-IV when it was given, otherwise the WASI-II 4-subtest FSIQ, otherwise the 2-subtest FSIQ. IQ < 90 is one of the ASD-Impaired criteria. The protocol reuses IQ from the referring study regardless of age.
No scores entered in SPINAPS REDCap yet.
T-scores (mean 50, SD 10) for the seven cognitive domains and the overall composite. The composite is one of the ASD-Impaired criteria (see Group Assignment Check).
No scores entered in SPINAPS REDCap yet.
Choosing which of four words best describes the mental state shown in 36 photographs of the eye region (total 0-36, higher = better).
No scores entered in SPINAPS REDCap yet.
40 faces, each happy, sad, angry, fearful or neutral: correct responses in total and per emotion (8 faces each).
No scores entered in SPINAPS REDCap yet.
Adaptive behaviour composites (standard scores, mean 100, SD 15): general adaptive composite (GAC) and the conceptual, social and practical domains. GAC < 90 is one of the ASD-Impaired criteria.
No scores entered in SPINAPS REDCap yet.

Participants whose PET Scan Information form is completed, by group: injected dose and mass, the arm used for the arterial line, samples collected, and whether the participant had side effects or complications from the arterial line or the scan. Technician notes are from the PET QC form (notes recorded for the scan or the blood draw). These are SPINAPS procedures, so there is no origin-study tab.
Specific activity (at synthesis and at injection) is not shown: the
REDCap fields have no units and the values recorded differ by orders of
magnitude, which suggests more than one unit is in use (see Data
Quality). Dose and mass values outside their valid range
(config/score_ranges.csv) are left out and listed under
Data Quality.

What is shown. Current psychotropic medications from
the SPINAPS Concomitant Medication Record (Psychotropic), for
participants whose form is completed. The protocol re-assesses
medications in SPINAPS even when they were recorded in the referring
study, so there is no origin-study tab. Medication names are free text;
config/med_classes.csv maps each name (including the
misspellings found) to a generic name and class. Entries with a
discontinuation date or marked as not continuing are left out. SSRI and
antipsychotic use are matching variables in the protocol.

Typed IDs to correct. IDs on the Shared Data
Availability form that are not exactly an origin record ID. Most were
corrected automatically for linking (the SOASD typo,
CMH for CMP in SPIN-ASD, hyphens, a trailing
. or /); some match no record. Please correct
them on the form so the links do not depend on these fixes.
