This report tracks recruitment, assessment completion and clinical scores for RMA01, a study of repetitive transcranial magnetic stimulation (rTMS) for depression in young adults with autism. REDCap data are pulled when the report is built; NIH Toolbox scores and MRI quality metrics come from local exports.

Test records, one duplicate record and one participant who was never allocated to treatment are left out of every summary. Score values that fail the range checks are left out and listed under Data Quality at the end.

For questions about the methods or code, contact Hassan Abdulrasul ().

Recruitment

Assessment Completion Status

Each heatmap shows one visit: only the forms given at that visit, and only participants who have reached it. Green = completed, gold = partly completed, red = recorded as not completed, grey = nothing entered. Hover over a cell for the recorded answer and the reason a form was not completed. Forms with an old and a new version (HRSD-17, ADOS-2, Pre- and Post-rTMS questionnaires) share one column showing the more complete of the two; the hover lists both. Self-report forms only have REDCap’s own status: “Incomplete” is shown as gold when the form has some answers and grey when it was never opened. Dates, rater and data-entry details, calculated scores and unticked checkboxes don’t count as answers.

Demographics

Baseline characteristics Started treatment: attended the first treatment session. Completed treatment: the completers used for the plots over time (12-weeks was captured by using RRS scores at 12 weeks), all of whom started treatment. Complete cohort: the same table for everyone consented. Everything is from the screening visit. Mean (SD) for scores, n (%) for counts; percentages are of participants with a value, and missing values are listed under each table.

WASI-II = Wechsler Abbreviated Scale of Intelligence. ADOS-2 = Autism Diagnostic Observation Schedule (calibrated severity 1-10, from the current form only). SRS-SF = Social Responsiveness Scale, short form. HRSD-17 = Hamilton Rating Scale for Depression (v2 form where both versions exist). QIDS-SR16 = Quick Inventory of Depressive Symptomatology, self-report, scored from its 16 items. MM-PHQ-9 = modified Patient Health Questionnaire-9. GAD-7 = Generalized Anxiety Disorder-7. BSS = Beck Scale for Suicide Ideation. CGI-S = Clinical Global Impression, severity, rated by the study clinician and separately by a research assistant (0 = not assessed is treated as missing). BDI-II = Beck Depression Inventory. GAF = Global Assessment of Functioning. C-SSRS = Columbia Suicide Severity Rating Scale (past-month ideation is asked only after a Yes to wish to be dead or suicidal thoughts; both No = no ideation). SBQ-R = Suicidal Behaviors Questionnaire-Revised (3-18). ATHF = Antidepressant Treatment History Form, covering the current depressive episode only; adequate trial = ATHF score of 3 or more (dose and duration), any trial type (medication, augmentation, ECT, rTMS, psychotherapy). The form records adequacy, not response, so trials are adequate, not necessarily failed. SCID = Structured Clinical Interview for DSM-5 (current = meets criteria now). Scores outside their valid range are left out (see Data Quality).
Women / men: sex assigned at birth. Gender diverse: a gender identity other than man or woman (trans man, trans woman, two-spirit, non-binary, other), or man / woman different from sex at birth; intersex and “do not know” are not counted. Race: self-reported (check all that apply), grouped as in the race plot; more than one group ticked is shown as such. Employment: full- or part-time paid work.
ADOS-2 classification: the current form offers Autism or Non-spectrum only; older forms recorded it as text. Previous suicide attempt: C-SSRS lifetime actual attempt (participants with no lifetime or past-month ideation count as no attempt). SCID depressive disorder is a single choice in REDCap, so MDD and PDD cannot both be recorded. Medications: psychotropic medication record at screening, drugs not marked as stopped; augmentation = with at least one antidepressant.
Not available in REDCap: age of onset, handedness, IDS-30, current episode duration.
Missing (started treatment / completed treatment): Race 1 / 0; Full-scale IQ (WASI-II) 4 / 3; ADOS-2 classification 8 / 6; ADOS-2 calibrated severity score 13 / 10; SRS-SF total, self-report 2 / 0; SRS-SF total, informant 2 / 0; Vineland adaptive behaviour composite 9 / 6; QIDS-SR16 2 / 0; MM-PHQ-9 13 / 9; GAD-7 5 / 2; CGI-S (clinician) 2 / 2; CGI-S (research assistant) 2 / 2; GAF 2 / 2; BSS (Beck Scale for Suicide Ideation) 5 / 3; C-SSRS most severe ideation, past month 3 / 2; C-SSRS ideation frequency, past month 3 / 2; Previous suicide attempt (C-SSRS, lifetime) 3 / 2; SBQ-R total 2 / 0; Major depressive disorder 2 / 1; Persistent depressive disorder 2 / 1; Any anxiety disorder 2 / 1; ATHF score 1 / 1; Adequate trials (ATHF score ≥ 3) 1 / 1; Electroconvulsive therapy 1 / 1; Psychotherapy 1 / 1; Benzodiazepine 1 / 1; Antidepressant 1 / 1; Antidepressant combination 1 / 1; Antipsychotic augmentation 1 / 1; Lithium augmentation 1 / 1.

Clinical Scores Over Time

Mixed Effects Model

Who is shown. Participants who completed treatment. Each coloured line is one participant; the boxplots below each plot show the spread at every timepoint with the mean as a black line.

The trend line is a linear mixed-effects model

score = intercept + slope × weeks since first treatment + participant offset + error
  • Why a mixed model and not an ordinary regression line. Each participant contributes up to ten scores, and scores from the same person are much more alike than scores from different people. The intraclass correlation (ICC) in each caption is that share: 0.13 to 0.73 here, so most of the spread in the plots is who the participant is, not when they were measured. An ordinary regression treats every score as a different person; it then overstates how precise the trend is, and participants with more visits pull the line towards themselves. The mixed model gives every participant their own starting level (the participant offset, a random intercept) and estimates the slope from how each person changes relative to themselves.
  • The slope is the average change per week across participants. Letting each person also have their own slope (a random slope) is not attempted: with 17 completers it is unlikely to be estimated reliably.
  • Note that the timepoints are not evenly spaced in time: the wait from screening to the first session ranges from 1 to 16 weeks, session 30 comes a median 6.1 weeks after the first session, and the 12-week follow-up a median 19 weeks after it. Time is measured from each participant’s first session (their earliest treatment date) using the assessment date of each score, so everyone is lined up on the treatment itself and screening falls at negative weeks. The intercept in the caption is the model’s score on the day of the first session.
  • Why the line looks bent. The x axis keeps the visits evenly spaced so every visit is readable. The trend is a straight line in real weeks, drawn at each timepoint’s median week, so it bends where visits are far apart in time (screening, follow-up). Hover over the black line to see the week of each point. The grey band is the 95% confidence interval of the average trend.

Hamilton Rating Scale for Depression

Beck Scale for Suicide Ideation

Modified Patient Health Questionnaire-9

Self-report, at screening, every fifth session and the three follow-up visits. The form has no date of its own, so time is taken from the visit date.

Beck Depression Inventory

Self-report, at screening, the lead-in visit two weeks later and the three follow-up visits (not at treatment sessions).

Clinical Global (severity) Impressions Scale

Clinician

Rated by the study clinician at screening and the three follow-up visits.

Research assistant

Rated by the research assistant at every assessment visit.

Clinical Global (Improvement) Impressions Scale

CGI-I is rated against the participant’s condition at the previous visit (1 = very much improved, 4 = no change, 7 = very much worse), so it is already a change score and is not asked at screening. There is no trend line: a participant rated 3 at every visit is improving each time, which a fitted line would show as flat.

Clinician

Rated by the study clinician at the three follow-up visits.

Research assistant

Rated by the research assistant at every assessment visit after screening.

MRI Quality (MRIQC)

T1-weighted structural

CNR (Contrast-to-Noise Ratio): Reflects GM/WM contrast.
Higher is better; typical range: 1.5–2.5.

SNR (Signal-to-Noise Ratio): Overall image clarity relative to background noise.
Typical range: 25–40.

INU Median (Bias Field Inhomogeneity): Measures intensity non-uniformity; closer to 1 is ideal.
Typical range: 0.95–1.1.

CJV (Coefficient of Joint Variation): GM/WM intensity overlap; lower is better.
Typical range: 0.5–0.9.

BOLD resting state

FD Mean (Framewise Displacement): Average head motion per volume.
<0.2 mm is ideal; 0.1–0.3 mm is typical.

FD Num: Number of volumes with FD > 0.2 mm.
Lower is better; <20 common.

tSNR (Temporal SNR): Stability of voxel signal across time.
Higher is better; typical range: 50–100.

DVARS (Standardized): Change in signal between volumes; higher = more motion.
Values near 1 are typical.

Data Quality

Nothing is changed in REDCap: every check below lists what to correct there. Score values that are outside their valid range, not a number, or a REDCap missing-data code are left out of all tables, plots and models. The other checks only list records for the team to review. Forms are checked at every heatmap visit (screening once, for all participants).
